Subject: no kidding - we need small crystals!
Hi everybody, Any suggestions regarding how to generate small!! crystals? Yes, in this case that is what we want While we can generate crystals of size 100-500 microns, this time we something in the range of .5-5 microns. Any ideas and suggestions would be greatly appreciated, Cheers Jens

Dear Jens,
It depends on the crystallization conditions. I am not sure from your
message whether you are looking for small crystals from any protein
(like lysozyme) or of a specific protein. If it is a specific protein
and if it crystallized in PEG conditions,you can increase the
concentration of PEG which might give you shower crystals of very
small size.

Try seeding so that you get 1000 crystals per drop for example. You know it's a pain to mount .5 micron crystals or do you want to suck them into a capillary ? You could also grind your big crystals and see if they still diffract. Smash up big Crystals?
There was a guy working on powder diffraction of protein samples (Lysozyme,
natch) in Birkbeck whilst I was doing my PhD and we all groaned when he
grew enormous crystals and then ground them up to collect powder data.

Hello, Definitely try microseeding: take a big crystal, put it in eppendorf with mother liquor ( the precipitant should be at a concentration higher than what you use to build the big crystals) and crash it by vortexing (put a small piece of plastic in the tube). If you use the vapor diffusion technique take a µl of this solution and mix it with your protein solution on a cover slip just as if you set a normal drop, and then seal the well.
